no everytime you do not use jailberak , you use jailbreak once, once you are past the jailbreak stage and can use iphoneinterface then you no longer need to use it. from my understand you only need to redo the services.plist once also, so once you upload it after you jailberak it and iphoneinterface it, that is done.
I mod my iphone regularly and i never put the services.plist during these mods , when i mod also i dont use jailbreak, only iphoneinterface. In windows (and i know you have osx but..) iphoneinterface is very tempremental and if you type stuff in wrong sometimes it crashes or lags etc. -- if it does just close it down and restart it back up and you should be back on track.
