Undoubtedly MobileSubstrate. Without it, most other tweaks in Cydia wouldn't be available.

Already voted, but some I like are

SBSettings (Simply works great. Works great even on my three year old iPhone 3G)
F.lux (Awesome for when I end up staying up late at night.)
WifiSMS (Looks good, to say the least)


Though, I do have some minor criticisms,

SBsettings is great, but can be a bit unresponsive and a little confusing with ordering the toggles to a certain way.
F.lux is awesome, but on my iPhone 3G it still has problems transitioning, so having two different options (one for day, one for night) completely grey screens my phone, forcing me to do a force shutdown
WifiSMS, looks great for composing long messages, (a tendency I have when I like to tell stories to my friends) but for whatever just plain out doesn't work on my iPhone. An update to fix that would help a LOT.

Overall, these are great. Assuming these all still work, these are going to be my go-to tweaks to get once I upgrade to something nicer in the future.

It's not any single tweak. It's the result you get when you combine any of the tweaks you want any way you want to make your phone how you want it.

Learn how to use a tethered JB, it's not bad! If you haven't been able to experience any iOS 5 JB tweaks then that's your fault.

why is BiteSMS not on the list?

#1 - BiteSMS (most used)
#2 - PhotoAlbums+ (most usefull)
#3 - SBSettings (most convenient)
#4 - Winterboard and/or Dreamboard
#5 - PandoraSkips (cant live without this one after using it)
#6 - iPicMyContacts (GOTTA have this)

those are my MUST have's... everything i could live without if i had to, but those are my main reasons ill jailbreak every iPhone ill ever have.

rainbow slide to unlock: changes the white flash that goes over slide to unlock and all other slide bars
