Well there's actually two separate ways posted here on the forums on how to make ringtones that are extremely easy.

If your iPhone is not jailbroken here's the tutorial for you;DIY: how to make a ringtone in under 60 seconds with nothing but iTunes

If your iPhone is jailbroken here is the tutorial for you;A "How To" Tutorial Guide for making your own Ringtones via iTunes

NOTE*** If your iPhone is jailbroken, your ringtones can be as long as you want them to be and if not, well then they can only be 40 seconds max (at least on iPhone 4 that is...30 seconds if 3gs or 3g I believe).

I've always used Garageband and it's a snap. Import your track, crop it to 40 sec and then export to itunes as a ringtone.
